ABSTRACT

Myocardial infarction (MI) is a common cardiovascular disease in clinical practice and one of the main causes of cardiovascular mortality. Its pathogenesis is complex and associated with oxidative stress reactions. Nuclear factor E2-related factor 2 (Nrf2) is a key factor in regulating oxidative stress reactions. It can regulate the expression of heme oxygenase-1 (HO-1), playing a role in maintaining the oxidative-reductive homeostasis in the body. During the course of MI, the biological activity and levels of Nrf2 and HO-1 decrease, leading to weakened tissue antioxidant and anti-inflammatory capabilities, endothelial damage in myocardial blood vessels, release of vascular cell adhesion factors, and impaired endothelial function. In recent years, many basic research studies have explored the role and mechanisms of traditional Chinese medicine (TCM) in treating MI by modulating the Nrf2/HO-1 signaling pathway. The results have indicated that the Nrf2/HO-1 signaling pathway is an important potential target for TCM in the treatment of MI. This article reviewed the mechanism of the Nrf2/HO-1 signaling pathway in MI and the research progress of TCM in targeting and regulating this pathway, aiming to provide a theoretical basis for the prevention and treatment of MI and further drug development.

ABSTRACT

OBJECTIVE:To establish a method for the content determination of saccharide in polysaccharides containing galact-uronic acid based on phenol-sulfuric acid method combined with correction factor method. METHODS:The determination condi-tion of phenol-sulfuric acid was optimized. A series of standard curves were drawn with glacturonic aid-glucose mixed control with different mass ratio. The content of saccharide in Codonopsis pilosula polysaccharides CPP1b samples containing galacturonic acid was determined. According to regression equation of galacturonic acid-glucose ratio of 0-100%,the correction factor was calculated by using C. pilosula polysaccharides CPP1b as the reference polysaccharide,and the results of content determination of saccharide were corrected. The rationality of this method was verified by using mixed monosaccharide control with same composition as C. pi-losula polysaccharides CPP1b. RESULTS:The correction factor of C. pilosula polysaccharide CPP1b to glucose was 3.33;in vali-dation test,the content of saccharide in mixed monosaccharide control with same composition as C. pilosula polysaccharides CPP1b was 103.47%. RSDs of precision and stability tests was <1%. The recoveries ranged 93.52%-107.35%(RSD=5.09%,n=6). CONCLUSIONS:The established method can accurately determine the content of saccharide in C. pilosula polysaccharides CPP1b containing galacturonic acid.

ABSTRACT

Objective To study the effects of different drying processes on the effective constituents in Wen Codonopsis pilosula (WPP) Decoction Pieces; To develop the optimized producing and preparing integration process based on fresh WPP.MethodsFresh WPP in harvest period was prepared respectively as follows:① Fresh WPP was dried to different percentage (30%–100%) of original moisture contents of crude drugs at 80℃ in oven, then sliced and dried at 50℃ to obtain eight decoction pieces of WPP (XⅠYP1–8).② The fresh WPP was baked to 50% of water content of crude drug under different temperatures (50–120℃), respectively, then sliced and dried at 50℃ to obtain eight decoction pieces of WPP (XⅡYP1–8).③ Dried WPP was moistened, sliced and naturally dried, then were renamed as traditional decoction pieces. The contents of lobetyolin and atractylenolideⅢ was determined by HPLC. The phenol-sulfuric acid and colorimetric method were applied respectively to detect contents of polysaccharide and total flavonoids. The contents of aqueous and alcoholic extracts were determined simultaneously. Results The contents of alcoholic extracts (55.36%), aqueous extracts (54.91%) and atractylenolideⅢ (10.95 μg/g) in XⅠYP3 pieces were higher than other decoction pieces.Conclusion The optimized process was that fresh WPP was baked to water content of 50% at 80℃, then sliced and dried. Compared with conventional preparing methods,the integration process was time-saving and effort-saving. Meanwhile, the prepared pieces have higher content of active ingredients.

ABSTRACT

Objective To observe the molecular mechanism of adaptive response of the kidney and skeleton and brain issues in the high altitude hypoxia; To discuss the unity of yin and yang oscillation relationship of kidney and brain marrow.Methods SPF KM mice were randomly divided into control group and model group according to random number table method. Mice in the model group were exposed to high altitude hypoxia cabin for successive 21 d. On the 22nd day, mice got out of the cabin and their body weight was measured, and then they were put to death through eyeball blood sampling. The activities of lactic LDH and Na+-K+-ATPase in brain tissue were detected by spectrophotometric colorimetry. The PFK activities of brain and skeletal muscle were detected by ELISA. Meanwhile the contents of EPO and EPOR in the kidney were measured by ELISA. The mRNA expressions of HIF-1α and AQP-4 in brain were assessed by RT-PCR. At the same time, the protein expressions of HIF-1α and AQP-1 in brain and the protein expression of Mb in skeletal muscle were detected by Western blot.Results Compared with the normal group, the LDH and PFK in brain tissue and the content of EPO in kidney tissue were all raised in the model group(P<0.05). Meanwhile the mRNA expressions of HIF-1α and AQP-4 and the protein expressions of HIF-1α and AQP-1 in brain were all increased in the mice from the model group; the activities of PFK and the protein expression of Mb in skeletal muscle were also raised in the model group. But the activity of Na+-K+-ATPase in brain tissue and the content of EPOR in kidney tissue both decreased in the model group (P<0.05,P<0.01).Conclusion Adaptive response and the unity of yin and yang oscillation relationship between kidney, skeleton and brain tissue happen in high altitude hypoxia.

ABSTRACT

Objective To observe the effects of volatile oil of Angelicae Sinensis Radix on blood pressure and related inflammatory cytokines of hypertensive rat models; To investigate the action mechanism of regulating the blood pressure.Methods Wistar male rats were fed withL-NNA to establish hypertensive rat models. Hypertensive rats were randomly divided into normal control group, model group, positive medicine group, volatile oil of Angelicae Sinensis Radix high- and low-dose groups, with 5 rats in each group. Each medication group was given relative medicine for gavage for 8 weeks. Non-invasive blood pressure measurement system was used to observe the systolic pressure of tail artery of rats before and after medication; the levels of ET-1, CRP, and VCAM-1 in serum were detected by the method of ELISA; RT-PCR was used to measure CRP mRNA of heart.Results The levels of systolic blood pressure, ET-1, VCAM-1, CRP and CRP mRNA in model group were significantly higher than those of normal group (P<0.05,P<0.01); Compared with model group, the levels of ET-1, VCAM-1, CRP and CRP mRNA in model group significantly decreased (P<0.05, P<0.01).Conclusion Volatile oil of Angelicae Sinensis Radix can inhibit vascular inflammatory response and then realize the effects of regulating blood pressure.

ABSTRACT

ObjectiveTo observe the effects of ultrafiltration extract from Angelica Sinensis Radix and Hedysari Radix on PC12 cell apoptosis induced by H2O2; To discuss its mechanism of action.Methods H2O2 was used in the incubation of PC12 cells to establish the oxidative damage nerve cell apoptosis model. The experiment was divided into normal control group, model group, and three different dosages (0.375, 0.75, 1.5 g/L) of ultra-filtration extracted from Angelica Sinensis Radix and Hedysari Radix after modeling for interference. Rate of cell apoptosis was detected by flow cytometry; mitochondrial membrane potential was measured by laser scanning confocal microscopy; the protein expressions of cleaved Caspase-3, Bax and Bcl-2 were detected by Western blot.Results Compared with the model group, rates of cell apoptosis in the different dosage groups decreased significantly; membrane potential of mitochondria increased; the protein expressions of cleaved Caspase-3 and Bax decreased; the expression of Bcl-2 increased; the ratio of Bcl-2 and Bax increased (P<0.05).Conclusion The ultra-filtration extracted from Angelica Sinensis Radix and Hedysari Radix can inhibit PC12 cell apoptosis induced by H2O2.

ABSTRACT

Objective To investigate the effects of ultra-filtration extract from the mixture of Angelicae Sinensis Radix and Hedysarum Polybotrys (UFE-AH) on the expressions of HSP70 and eNOS in H2O2-induced endothelial cell apoptosis. Methods H2O2 induced ECV-304 cell apoptosis to prepare models. The experiment was divided into normal control group, model group, simple medicine group, medicine intervention group, and all treatment groups received relevant medicine for intervention. Flow cytometry (FCM) was used to detect apoptosis and concentration of intracellular Ca2+;RT-PCR was used to detect the mRNA expression of HSP70 and eNOS;Western blot was used to detect the expression of HSP70 protein;Nitrale reduetase and spectrophotometric method were employed to detect the content of NO. Results Compared with normal control group, cell apoptosis rate, concentration of intracellular Ca2+, and expression of HSP70 increased significantly in model group (P<0.05);gene expression of eNOS mRNA and content of NO decreased (P<0.05). Compared with model group, cell apoptosis rate and concentration of intracellular Ca2+dropped in medicine intervention group (P<0.05);expressions of HSP70, eNOS mRNA and content of NO increased (P<0.05). Conclusion UFE-AH can confront H2O2-induced cell apoptosis H2O2 of ECV-304 human umbilical vein endothelial by increasing the expressions of HSP70 and eNOS and content of NO, and reducing the intracellular calcium overload.

ABSTRACT

Objective To explore the intervention effect of active fraction of Angelica Sinensis Radix in mice under high altitude hypoxia condition. Methods Totally 72 healthy SPF mice were randomly divided into control group (K), model group (M), Rhodiola rosea group, and active fraction of Angelica Sinensis Radix groups (B, C, X). The mice were administerted corresponding treatment by gavage for 21 days. Control mice were given normal saline in same volume. From the 8th day, all mice excepted control mice were exposed to high altitude hypoxia cabin after 0.5 hour gavage treament. On the 22nd day, after got out of the cabin and their body weight were measured, mice were put to death through eyeball blood sampling to prepare splenic lymphocyte suspension. The proliferation and transformation capacities of lymphocyte cell and killing activity of NK cells were detected by MTT. The content of IL-2 in the serum of mice in each group were detected by ELISA. Results Compared with the control group, the body weight of mice, the proliferation and transformation capacities of lymphocyte cell, the killing activity of NK cells, and the content of IL-2 were all significantly decreased (P<0.05, P<0.01). Experiment tests showed that the proliferation and transformation abilities of lymphocyte cell and the killing activity of NK cells were all increased in the mice of group B, C, and X compared with those of the model group (P<0.05, P<0.01). The stimulate index of lymphocyte cell was raised after X intervention compared with those of the model group (P<0.05). The content of IL-2 in the serum was enhanced after intervention of active fraction C and X of Angelica Sinensis Radix compared with those of the model group (P<0.05, P<0.01). Conclusion Active fraction of Angelica Sinensis Radix shows increasing immunological function of mice exposed to hypoxia.

ABSTRACT

Objective To investigate the genetic relationships of Astragalus membranaceus and Codonopsis pilosula from the different cultivars (varieties). Methods Twenty-five samples of A. membranaceus and seventeen samples of C. Pilosula were amplified by ISSR-PCR. Genetic diversity and genetic distance were analyzed by Popgen32. UPGMA dendrogram relationship was clustered by NTSYS. Results Totally 62 and 100 loci were detected respectively from A. membranaceus and C. pilosula. All percentages of polymorphic loci (PPL) were 100%. Shannon’s information indexes (I) were 0.537 6 and 0.472 7. Nei’s genetic diversity indexes (H) were 0.361 3 and 0.307 4. A. membranaceus was divided into two groups in genetic distance 0.59, and C. pilosula was alike in 0.50. Conclusion Different cultivars (varieties) of A. membranaceus and C. pilosula had rich genetic diversities among species, and had large genetic differences.

ABSTRACT

This study was designed to investigate the impact of ultra-filtration extract mixture from Astragals mongholicus (UEMAM) o radiosensitivity of H22 ascitic tumor in mice to 12C6+ ions radiation. The H22 ascitic tumor model was established in mice by intraperitoneal injection of 0.2 mL H22 ascitic cells. The animals were subsequently divided into 4 groups randomly, treated with normal saline, UEMAM, heavy ion beam radiotherapy and UEMAM plus heavy ion beam radiotherapy, respectively. The body weights, abdomen circumference of the mice were measured and the mouse behavior was monitored every day; survival time was recorded to evaluate life extension effect; flow cytometry technique was used to detect H22 cell apoptosis and cell cycle; protein levels of p53, Bax, Bcl-2 and cleaved Caspase-3 were analyzed by Western blot; the single cell gel electrophoresis was used to detect the level of deoxyribonucleic acid damage (DNA damage). The results suggest that UEMAM significantly increased survival time, and decreased body weights and abdomen circumference over the saline control group. The treatment increased cell apoptosis, cycle arrest and DNA damage compared to the saline control group. UEMAM significantly enhanced the therapeutic effect of heavy ion beam radiation in survival time, and decreased body weights and abdomen circumference in the tumor-baring mice. The combination increased cell apoptosis, cycle arrest and DNA damage compared to the radiotherapy group. The results of Western blot suggest that the treatment significantly enhanced p53-induced apoptotic signals. The experiment discovered that UEMAM could improve radiosensitivity of H22 ascitic tumor through activation of p53-mediated apoptotic signal pathway.

ABSTRACT

Objective: To explore the relationship between ambulatory arterial stiffness indexes (AASI), AASI-blood pressure variability (AASI-BPVR) and left ventricular mass index (LVMI) left atrium diameter (LAD) in patients with hypertension. Methods: A total of 286 hypertensive patients were enrolled in this study. The AASI, AASI-BPVR were calculated from 24-hour ambulatory blood pressure monitoring. Left ventricular internal dimension in diastole (LVIDd), interventricular septal thickness in diastole (IVSd), posterior wall thickness in diastole (PWd), LAD were detected by echocardiography and LVMI, relative wall thickness (RWT) were calculated. The fasting blood glucose, blood lipids were examined. According to 24 h AASI, the patients were divided into 2 groups, Group A, the patients with AASI > 0.51, n=133 and Group B, the patients with AASI ≤ 0.51,n=153. Pearson and multi regression analysis were conducted to analyze the relevant correlations. Results: Group A had increased LVMI than that in Group B,P Conclusion: AASI and AASI-BPVR were not the independent factors for left ventricular hypertrophy and left atrial enlargement, therefore, they were not the predictors for cardiac damage in patients with hypertension at present time.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate diseases occurring in Rheum palmatum in Gansu province and identify pathogen.</p><p><b>METHOD</b>Field survey in the main R. palmatum producing area and the pathogens were identified on the base of morphology.</p><p><b>RESULT</b>Eight fungal diseases have been found after disease surveys on R. palmatum in Gansu province from 2004 to 2007. They were leaf smut (Thecaphora schwarzmaniana), leaf spot (Septoria sp.), rust (Puccinia rheipalmati), ring spot (Ascochyta rhei), powdery mildew (Erysiphe polygoni.), grey mold (Botrytis sp.), root rot(Fusarium oxyxporium) and a virus disease.</p><p><b>CONCLUSION</b>Leaf smut, a severe disease with the incidence of 14%-26%, is the urgent problem waiting to be solved effectively. Powdery mildew and grey mold were reported for the first time in China.</p>

ABSTRACT

Objective To isolate and purify polysaccharide from Angelica sinensis by ultrafiltration technology. Methods Extract of polysaccharide of Angelica sinensis was ultrafiltrated with different aperture membrane, the separation parameters on membrane separation efficiency of polysaccharide of Angelica sinensis were optimized by and orthogonal test. Results Polysaccharide of Angelica sinensis was separated with 200, 100, 50, 20 kD membrane. The contents of polysaccharide was 30.47% before ultrafiltration. The main component of Angelica sinensis polysaccharide was the fraction with 200 kD (65.42%). The optimum condition of ultrafiltration was obtained as follow:one times of extract volume, 35 ℃, 0.3 MPa. Conclusion The technology was simple and available, and the content of Angelica sinensis polysaccharide was higher.

Object To set up an identified standard on the level of molecule through measuring the internal transcribed spacer regions of the rRNA gene in Radix Angelicae Sinensis Methods To extract DNA from the seeds of Radix Angelicae Sinensis by conventional method, and use composed peculiar primer amplify with the internal transcribed spacer regions of the rRNA gene, to measure the base sequence of the amplified products Results It is proved by agar sugar gel electrophoresis that the PCR amplified products of the internal transcribed spacer regions of the rRNA gene exist The base sequence of the seeds of Radix Angelicae Sinensis internal transcribed spacer regions of the rRNA gene was measured Conclusion To measure the base sequence of internal transcribed spacer regions of the rRNA gene is another effective method to identify the vegetal traditional Chinese medicine on the molecular level

ABSTRACT

Objective To investigate whether the administration of the ultra-filtration extract mixture from Angelica sinensis and Hedysarum polybotrys is able to protect cardiomyocytes from oxidative injury of rats induced by H2O2 and its potential mechanism. Methods Myocardial cells from 2—3 d neonatal rats were cultured in DF medium and the cellular injury was induced by H2O2. The ultra-filtration extract mixture from A. sinensis and H. polybotrys was given in three doses of 3.75,7.5,and 15 mg/mL. Morphological changes of cardiomyocytes were observed by microscope. Survival rate of myocardial cells was assessed using MTT. The cardiomyocyte damages were estimated by detecting lactate dehydrogenase (LDH) and creatine kinase (CK) releases in the medium,superoxide dismutase (SOD) activities and intracellular malondialdehyde (MDA),and myeloperoxidase (MPO) contents. The levels of caspase-3 and hsp70 expression in cardiomyocytes were measured by RT-PCR. Results The ultra-filtration extract mixture could protect the cardiomyocytes from H2O2 injury in a dose-dependent manner (3.75,7.5,and 15 mg/mL). The ultra-filtration extract mixture could significantly decrease LDH and CK leakages and intracellular MDA and MPO contents,increase SOD activity,upregulate hsp70 expression,and downregulate caspase-3 expression. Conclusion The ultra-filtration extract mixture has protection on cardiomyocytes injured by H2O2 through improving cell antioxidant ability,upregulating hsp70 expression,and inhibiting caspase-3 activity.

AIM: To study the effect of three kinds of drying methods on quality of Radix Angelicae Sinensis. METHODS: TLC, HPLC determination of volatile Oil and ethanol-extra ctives were used. RESULTS: The constitutents in three kinds of dried samples are the same, the contents of vo latile oil and ferulic acid have evident difference in three kinds of aried samp les. The content of fumed sample is the highest. CONCLUSION: The fuming method is the best drying one.
